I have mobile USB Hard Drive, i always take around with me but i'm afraid that my co-worker might take it to steal my files or important documents.

One possibility is using whole-disk encryption, and there are several free alternatives such as TrueCrypt. Pay special attention to the FAQs and the warnings, though, because you can lose access to the files if you are not careful.

Unless you have formatted the external drive as NTFS, you cannot use the built-in encryption provided by Windows, assuming you are using Windows.
